Four numbers that are commonly confused
A peptide report may contain chromatographic area purity, measured peptide content, total dry or lyophilized mass, and expected molecular mass. These numbers have different units and purposes. Area purity is usually a percentage of integrated chromatographic signal. Content is commonly expressed as mass per vial.
Total powder mass includes all solids and retained volatile material. Molecular mass describes one molecule, not the quantity in a container.
Why high area purity can coexist with a content difference
A sample can have one dominant chromatographic component yet contain less or more target material than a nominal fill claim. This can result from filling variation, moisture, counterion form, reference-standard assignment, sample preparation, or the quantitative method.
The purity chromatogram may still look clean because it measures relative signal among detected components rather than the absolute amount placed in the vial.
What to request from a supplier
Ask which method supports identity, which method supports purity, whether content was measured, the basis used for quantitative calculation, the tested batch, and whether raw chromatographic or spectral evidence is available. A clear supplier should not use one percentage to imply every quality attribute.
